Researchers have identified a signaling pathway that can regenerate damaged tissue in the airways and pancreas of mice, offering potential benefits for individuals with respiratory illnesses and metabolic syndrome. The study, conducted by Stanford Medicine researchers and their colleagues, found that a rare cell type in the lung lining is crucial for repairing damage from toxins like wildfire smoke or respiratory viruses. A similar process occurs in the pancreas, where neuroendocrine cells protect insulin-producing pancreatic islet cells.
The research involved treating mice airways with an experimental drug to activate the repair pathway, which protected them from damage after infections such as influenza or COVID-19. Blocking this pathway resulted in more severe airway damage. Researchers believe activating this signaling pathway in humans could help prevent permanent lung damage and aid those with metabolic syndrome from progressing to diabetes.
“This whole signaling cascade both protects and regenerates vulnerable cells in the airway and the pancreas,” said Philip Beachy, PhD, professor of urology and developmental biology. “If this circuit is disrupted, the damage is much worse – specialized airway cells are lost, and the stem cells can’t divide to repair the damage. We think it’s likely to be important in many other tissues in the body.”
The study was published online on June 9 in Cell. Research scientist William Kong, PhD, is the first author of the study.
Neuroendocrine cells make up less than 1% of airway lining cells but play an essential role in immune response modulation. The study highlighted how these solitary neuroendocrine cells function within epithelial tissues like airways.
Beachy’s lab has long studied Hedgehog proteins’ role since identifying them initially in fruit flies. These proteins are vital for embryo development but also assist tissue rejuvenation. Previous work indicated Sonic hedgehog protein involvement in bladder lining regeneration after bacterial infection.
In this study, Desert hedgehog protein presence was detected using bulk RNA sequencing techniques on tracheal cells; its activation led to Gli1 protein production essential for epithelial repair processes following toxin exposure.
“At each stage, the signal is amplified until the entire trachea is impacted,” Beachy explained regarding rapid protective responses triggered upon detecting epithelial damage.
